Designing tiny molecules that glow in water to shed light on biological processes

ScienceDaily (Aug. 15, 2012) ? University of Miami scientists have developed a way to switch fluorescent molecules on and off within aqueous environments, by strategically trapping the molecules inside water-soluble particles and controlling them with ultraviolet light. The new system can be used to develop better fluorescent probes for biomedical research.

Previous studies have used water-soluble particles to bring organic molecules into water. What is novel about this system is the use of a photoswitching mechanism in combination with these particles.

The findings published online by Chemistry-A European Journal, describe the creation of a fluorescent photoswitchable system that is more efficient than current technologies, says Francisco Raymo, professor of chemistry at the UM College of Arts and Sciences and principal investigator of this study.

"Finding a way to switch fluorescence inside cells is one of the main challenges in the development of fluorescent probes for bioimaging applications," Raymo says. "Our fluorescent switches can be operated in water efficiently, offering the opportunity to image biological samples with resolution at the nanometer level."

Fluorescent molecules are not water soluble; therefore Raymo and his team created their system by embedding fluorescent molecules in synthetic water-soluble nanoparticles called polymers that serve as transport vehicles into living cells. Once inside the cell, the fluorescence of the molecules trapped within the nanoparticles can be turned on and off under optical control.

"The polymers can preserve the properties of the fluorescent molecules and at the same time assist the transfer of the molecules into water," Raymo says. "It's a bit like having a fish in a bowl, so the fish can carry on with its activities in the bowl and the whole bowl can be transferred into a different environment."

The new system is faster and more stable than current methods. The fluorescent molecules glow when exposed simultaneously to ultraviolet and visible light and revert back to their original non-luminous state in less than 10 microseconds after the ultraviolet light is removed.

By using engineered synthetic molecules, the new system is able to overcome the natural wear down process that organic molecules are subject to when exposed to ultraviolet light.

"The system can be switched back and forth between the fluorescent and non-fluorescent states for hundreds of cycles, without sign of degradation," Raymo says.

The surface of the system can be customize to help it attach to specific molecules of interests, thus allowing researchers to visualize structures and activity within cells, in real time, with a resolution that would otherwise be impossible to achieve.

Raymo and his team will continue improving the properties of the molecules for future biomedical applications. The study is titled "Fast Fluorescence Switching within Hydrophilic Supramolecular Assemblies" Co-authors are Janet Cusido, Mutlu Battal, Erhan Deniz and Ibrahim Yildiz,Ph.D., students in the Department of Chemistry at UM; and Salvatore Sortino, associate professor of chemistry in the Department of Drug Sciences, University of Catania, Italy. The research was supported by the National Science Foundation.

According to research, pregnant women who are mindful about their health as well as their baby?s health during pregnancy tend to produce healthier and fitter babies. Women who are less mindful about what they eat and what they do, tend to expose their babies to higher risk of suffering from birth defects and other undesirable consequences. So what are the things that you should be mindful about during pregnancy?

Alcohol Consumption
Consumption of large amount of alcohol imposes harm to you and your baby. BOA chairman Moynihan to step down

LONDON (Reuters) - British Olympic Association (BOA) chairman Colin Moynihan will step down from his post later this year to give his replacement time to settle in before the 2014 Sochi Winter Olympics, the 1980 rowing silver medalist said on Monday.

Moynihan, who oversaw the best British medals haul in a century at the Games which finished on Sunday, has been in the role since 2005 and will hand over to a new head later this year, with a vote likely in November, a BOA spokeswoman said.

"He or she must be integrally involved in the preparations for Sochi," Moynihan wrote in a letter to the BOA and the National Olympic Committee.

The former Sports Minister will focus on his family and his energy sector business interests but will maintain involvement with the BOA to help choose his successor.

British athletes bagged 65 medals in total, including 29 golds, to finish third in the London table behind the United States and China, a far cry from the one gold medal at the 1996 Atlanta Games.

Moynihan announced he would also be stepping down as chairman of British Ski & Snowboard (BSS).

"BSS has been transformed since the dark days of the collapse of its previous governing body into administration on the very eve of the Vancouver Games," said Moynihan.

Britain won one gold medal in Vancouver in 2010 and the BSS is now backed by more than two million pounds ($3.14 million) raised in private-sector sponsorship and donations as they seek more success at the Winter Olympics.

Moynihan said he hoped sport would become more of a priority in schools after Britain's showing at the London Games.

"I believe that we need to review and where relevant re-think government sports policy in order to translate the inspirational effect of the Games directly into participation.

"The new requirement for primary schools to provide competitive sport is a step in the right direction. ($1 = 0.6373 British pounds)

A technique known as photoacoustics can find some forms of melanoma even if only a few cancerous cells exist, but a recent study found the technique is limited in its ability to identify other types of cancer. U. MISSOURI (US) ? Attaching ?enhancers? to cancer cells could help photoacoustic scans, which use lasers to detect early signs of melanoma, identify other types of cancer.

?Eventually, a photoacoustic scan could become a routine part of a medical exam,? says Luis Polo-Parada, assistant professor of pharmacology and physiology at the University of Missouri. ?The technique doesn?t use X-rays like current methods of looking for cancer. It could also allow for much earlier detection of cancer.

?Now, a cancerous growth is undetectable until it reaches approximately one cubic centimeter in size. Photoacoustics could potentially find cancerous growths of only a few cells. Unfortunately, our research shows that, besides some cases of melanoma, the diagnostic use of photoacoustics still has major limitations. To overcome this problem, the use of photoacoustic enhancers like gold, carbon nanotubes, or dyed nanoparticles is needed.?

Photoacoustics uses pulses of laser light to heat cells for a fraction of a second. When the cells become hot they emit a tiny sound. Extremely sensitive microphones can hear those sounds. The strength of the sound depends on how much laser light is absorbed.

Since darker objects absorb more light they also emit more sound and can be found using photoacoustics.

?Some melanoma can be found by photoacoustics because the cells contain large quantities of melanin, a dark pigment,? Polo-Parada says. ?Other cancers don?t have that much pigmentation; hence, they don?t stand out as much in photoacoustic scans. This is where enhancers may be able to help by labeling cancer cells and making them stand out in a scan.?

Polo-Parada in collaboration with Gerardo Gutierrez-Juarez, a researcher from the University of Guanajuato, Mexico, found that out of seven types of cancer cells, only one type of melanoma was dark enough to produce a sound strong enough to be distinguishable from the rest. Their findings are published in the journal AIP Advances.

The photoacoustic technique holds promise in the fight against cancer, notes Polo-Parada, but it is too soon to say exactly when the public will benefit. Eventually, other diseases that cause changes in the coloration of cellular tissue, such as malaria, could be found by photoacoustics.
